This innovation represents the development of an advanced drug delivery system for leukemia therapy using cyclodextrin-based nanosponges synthesized by a solvent-free approach. The system is based on cyclodextrins, cyclic oligosaccharides with a hydrophobic cavity that form inclusion complexes with hydrophobic drug molecules, while their hydrophilic outer surface enhances solubility and improves the bioavailability of the encapsulated compound. Crosslinking of cyclodextrin units forms a three-dimensional porous network with numerous binding sites, enabling higher encapsulation efficiency and precise control of drug release. The synthesis is carried out using mechanochemical and melt methods with crosslinkers, without the use of solvents, ensuring a sustainable process. The developed system containing dasatinib shows prolonged and controlled release compared to a commercial immediate-release dosage form, with a 30% increase in the final fraction released and complete release over an extended period. This enables more stable drug concentrations, reduces peak-related adverse effects, and allows for less frequent dosing.
